I want to build 2 Domino servers, of which one will be the interface to Web users and the other one will be the core server (will replicate with other additional servers and AS/400) as well as run DECS, ODBC etc. Do I have to build 2 first servers or one will be the additional?

It really depends, but the simplest way to do this is one "1st" server, your administrative server, and one additional server. This way you only have one domain to administer, and replication, mail routing, etc., will all take place simply, rather than needing cross-certificates and domain documents.
