What is the best method of getting data from a COM port (or RS232&TCP/IP)?
I haven't done this, and I wouldn't try to do it in LotusScript. Since serial input is an event-driven process, you need to use a language that lets you insert your own code as the receiver for those events.
It's probably easier to write an outside monitor program (written in C or C++, perhaps) that reads the information and stores it in text files that your LotusScript code can pick up when it's ready.
If you're brave, you could try using the Windows APIs from LotusScript, as was done here for writing to the COM port.
